between genistein group and normoxic group(P0.05). The percentage of muscularized arteries in
small pulmonary vessels of genistein group was significantly less than that in hypoxic group
(P<0.01). Genistein could directly reduce vascular smooth cell proliferation induced by hypoxia.
Conclusion: These findings indicated that genistein supplementation can prevent chronic hypoxia
induced pulmonary vascular structural remodeling. The inhibition of genistein on hypoxia
induced vascular SMC proliferation may be one of the important preventive mechanisms.

To identify which isoform of protein kinase C (PKCs) may play a key role in the
development of cerebral hypoxic preconditioning, the biochemistry technques of SDS-PAGE,
Western blot and Gel Doc imagine system were applied to semi-quantitatively analyze the effect
of repetitive hypoxic exposure on the level of PKCs membrane translocation and protein
expression in hippocampi and cortex of mice. Experiments were carried out in accordance with
the National Institutes of Health guide for the care and use of laboratory animals. The hypoxic
preconditioned mice model was adapted with minor modification from our previous report. In
brief, healthy adult BALB/C mice weighing 18~20g regardless of sex were randomly divided into
5 groups: control (H0), hypoxic control (H1, hypoxic exposure once), and hypoxic
preconditioned group (H2~H4, repetitive hypoxic exposure for 2~4 times respectively). We found
that the level of conventional PKC, and novel PKC's membrane translocation, which was
regarded as an indicator of PKCs activation, were increased significantly (*p<0.05, n=6 for each
group) in response to the repetitive hypoxic exposure both in hippocampi and cortex of mice, but
no significant changes in membrane translocation of the rest 8 isoforms of conventional PKCs,
novel PKCs and atypical PKCs were found. In addition, there were also no significant changes in
protein expression level of all 11 isoforms of PKCs both in hippocampi and cortex. These results
suggested that the activation of conventional PKCb_and g, and novel PKCe might play an
important role in the development of cerebral hypoxic preconditioning.

Objective: To examine the effect of hypoxia and cobalt chloride (CoCl2) on the proliferation
of hMSCs in vitro. Methods: hMSCs were isolated from human aborted fetus bone marrow (with
permission of local authority and the family) through density gradient percoll (1.073g/ml) and
expanded in vitro. The thirteenth passaged hMSCs were seeded at 5_105/60-mm tissue culture
dish and immediately divided into five equal groups as follows: 3%O2, 10%O2, 100uM CoCl2,
200uM CoCl2 and control group in 20%O2. The number of hMSCs were determined by
haemacytometer count and cell proliferative index were measured by flow cytometer. Results:
The number and proliferative indexes of hMSCs were higher for cells treated with 100_M CoCl2
or 200_M CoCl2 or maintained in low oxygen than for those of control group. The differences
between experimental and control groups are of statistical significance (P<0.001). These data
show that hypoxia can promote the proliferation of hMSCs in vitro. Conclusions: Low oxygen
and CoCl2 could be useful tools to expand hMSCs in vitro for clinic use. The work was supported
