Objective: to determine the sequence structure of globin cDNA in Tibetans living at high
altitude. Subjects and methods: total RNA was isolated from human bone marrow samples from
three Tibetans who live on the Qinghai-Tibet plateau. cDNA fragments coding for alpha and beta
genes of human hemoglobin were obtained through RT-PCR and were ligated to plasmid pGEM-
T, and then the ligation liquid was transformed (?transferred) to Escherichia coli and cloned.
DNA Sequence was compared with NCBI data using BLAST method. Results: the sequence of
globin cDNA in Tibetans was the same as the registered one in the NCBI, and no mutation points
were found in Tibetan globin coding gene. Conclusion: At the mRNA level, no unique sequence
was found in Tibetan globin coding genes.

Objective: To determine the changes in serum troponin (cTn-t) in patients with high altitude
pulmonary edema (HAPE) who had received nitric oxide treatment. Methods:
Radioimmunoassay was adopted to determine the amounts of serum cTnT-T in 21 HAPE patients
before and after nitric oxide treatment. All the HAPE patients inspired air containing 10 ppm NO
twice a day for 1 hour at a flow rate of 3 liters per minute. Blood oxygen saturation (SaO2) was
detected from the finger. After each NO treatment, the patients who still showed symptoms of
HAPE continued to breathe in low flux of oxygen. Students' t-test was used to analyze the
differences. Results: Among 21 HAPE patients, 6 subjects were cured in 2 days, 7 in 3 days, 5 in
4 days, and 3 in 5 days. 9 patients were cured by simply NO inhalation and 12 patients were
cured by NO plus oxygen supplementation. The serum level of cTnT-T in these patients was
0.2284±0.0502 ng/ml and 0.0443±0.0133 ng/ml before and after the nitric oxide treatment. The
finger blood oxygen saturation in these patients was 0.6±0.1 and 0.85±0.06 correspondingly,
which is statistically different (P<0.01). Conclusion: The changie in serum cTn-T indicated that
HAPE patients had suffered myocardial damage. Nitric Oxide or nitric oxide plus oxygen has
been proven to be an effective therapeutic in the control of severe HAPE.

To observe the ultrastructural changes of pituitary ACTH cells in rats after exposure to
different altitudes and to investigate the role of ACTH in hypoxic stress, including the
relationship between hypoxia and the endocrine function of pituitary ACTH cells in rats.
Methods: Wistar rats were divided into three groups and exposeed to either 1700m, 3100m, or
4050m for 12 days. Changes inpituitary ACTH cells of the rats were observed using
immunohistochemical and imaging methods. Results: Imaging analysis of intracellular hormone
content in the 1700m group showed that there were statistical significances compared with the
3100 m or 4050 m groups (P<0.05), but no statistical significance between the 3100 and 4050 m
groups. Conclusion: Hypoxia has some effects on the endocrine function of pituitary ACTH cells
in rats. ACTH may perhaps be a key physiologic and pathophysiologic factor involved in the
stress reaction to hypoxia in high altitude.

The objective of this study is to research the effect of high altitude on neurobehavioral
function in automobile drivers at altitude. Methods: Forty-two healthy drivers living at different
elevations (2260m~, 3000m, 3600m~, 3800m, 4000m~, 5000m) were tested using the NCTB
(Neurobehavioral Core Test Battery) developed by the WHO. The Neurobehavioral Core Test
