Developing novel proteins
There is a growing need for new or improved proteins for vaccines and
therapeutic application (e. g. proteins as antidote to C/B warfare agents).
The unique expertise and proprietary technologies of Protéus dramatically
accelerate the development of novel proteins.
Protéus' L-ShufflingTM-based directed evolution approach offers a number
of opportunities to create novel
antibodies, antigens, enzymes
and
other
active proteins
with new and/or improved characteristics such as affinity,
specific activity and half-lives etc...
L-ShufflingTM
is a proprietary directed evolution technology developed and
patented by Protéus to provide significant improvements over previous
gene shuffling techniques. Improvements include :
Ø High proportion of functional variants ;
Ø Engineering of small and large-sized genes ;
Ø High fidelity recombination steps ;
Ø Shuffling of genes featuring either low or high homology ;
Ø Choice of recombining entire genes or protecting certain regions.

DNA/RNA-based assays
Protéus has developed customized protocols for the extracting genetic
material from a variety of samples such as air, soil, sporulated bacteria or
body fluids and for eliminating of interfering compounds. Real-time
amplification techniques are routinely used to develop assays for the
detection, identification and quantification of pathogenic targets. Primers,
probes and protocols are designed to achieve a perfect fit with the required
speed, specificity, sensitivity, automation and ease of use.
Protein-based assays
Functional analysis of proteins is a key to researching immune response
against invading pathogens, or to assessing their sensitivity (or resistance)
to antibiotics or antiviral drugs. The proprietary
cell-free in vitro
expression technology PhenomicsTM
is a powerful tool for developing
diagnosis tests for

Ø Global activity detection (e.g. HIV protease activity) ;
Ø Viral or microbial population profiling (e.g. antibiotic or protease
resistance).

Developing novel enzymes for neutralizing
C/B weapons and for bioremediation
Enzymes are potent catalysts that can degrade, neutralize or lower the
toxicity of virtually any chemicals. Since inception, Protéus has been
developing enzymes that are active on a wide variety of industrial
chemicals.
Extremely robust enzymes
with unprecedented half-lives
under extreme conditions of temperature or pH have been successfully
developed.
Taking advantage of its unique access to biodiversity from extreme
environments, of its proprietary L-ShufflingTM-based directed evolution
platform and its know how and capabilities in scaling up enzyme production,
Protéus is ideally positioned to develop novel enzymes that will
neutralize
chemical or biological warfare agents.
Such enzymes can be
incorporated in products for use by military and civilian first responders,
such as firefighters, police and hazardous material response teams. The
enzymes have the additional benefit of being
non-corrosive
and
environmentally friendly
.
Enzyme-based decontamination technology may also be used in
bioremediation of industrial and agricultural waste sites and for the
destruction of abandoned chemical warfare agents stockpiles.
8,100 wells biochip
Within the frame of a collaboration with CEA-
LETI, (Commissariat à l'Energie Atomique,
France) Protéus has developed a wide range
of 3D biochips, allowing to perform any liquid
phase protocol (e.g. nucleic acid
amplifications, enzymatic activity tests, etc.)
under a miniaturized format : 16, 25, 100 and
8,100 wells formats are available. A
dedicated reader has also been developed for
real time kinetic analysis of the biochips
Scientists in the bioprocess pilot lab at
Protéus preparing a process book for the
industrial scale up of an enzyme.
Protéus has extensive experience in
developing novel proteins. Facilities include
clean rooms and a BL-3-safety level lab.
