Benefits of L-ShufflingTM
Based on recombination by ligation of gene fragments in the
presence of suitable template, L-ShufflingTM offers several
competitive advantages including :
Easy recombination of both small and large genes :
(L-ShufflingTM" refers to Large-ShufflingTM).
The
high fidelity
recombination generates a very
high
proportion of functional variants. No need for
screening millions of clones for identifying
valuable hits. Most experiments do not require
more than 1,000 clones to be screened.
Versatility : control over location and frequency of
recombination. L-ShufflingTM enables to shuffle the
entire gene sequences or leave intact defined regions.
It enables both controlled and fully randomized
recombination.
Versatility : recombination of both high and low
homology genes
Each round of L-ShufflingTM generates new variants which are
then screened for selecting the best ones with the most
improved performances. PhenomicsTM, the proprietary in vitro
cell-free expression system developed by Protéus is

using gene-shuffling
L-ShufflingTM of TNF-R1 variants : Valuable variants have
been
detected
within
only
one
round
of
L-ShufflingTM after
screening less than 600 clones
.
Approx. two thirds of the variants were active. The best
variants exhibit more than three fold improved binding
efficacy vs. the wild type (WT) gene. Variants have been
expressed using PhenomicsTM in 96-well MTPs
