To determine the CNA status of these
BSE-susceptible
cohorts,
fifteen
unrelated cohorts from German BSE
cases were tested. Figure 1 shows that
>33% of individuals in all 15 cohorts
demonstrated detectable CNA. In
addition, four of four sera from
confirmed PrP
res
positive BSE cows
showed positive results. These results
are statistically significant (p<0.001); if
calculated separately, every individual
cohort group has a significantly
(p<0.001) increased reactivity compared
to the healthy controls.
Normal Serum Samples
908 serum samples from known PrP
res
-
negative cattle were obtained from a
German slaughterhouse. CNA were
extracted and tested. Table 2 shows the
number of reactive samples. All animals
were tested with an approved prion test
for the presence of PrP
res
in the brain
stem. In each test run, an amplification
control, extraction control and negative
controls were run together with non-
template controls. In addition, a Cohort
Control panel was included in the runs.
The panel contained 23 GLT repeatedly
reactives and 12 GLT non-reactives.
Samples were blinded and run in the
GLT. Results were tabulated and the
code broken at the end of each working
day and the daily runs accepted if
controls were correctly identified. The
results of this study are shown in Table
2.
