New Ni Sepharose High Performance offers very high binding capacity
New affinity medium Ni SepharoseTM High Performance is optimized for the purification of His-tagged proteins. It provides
scientists in industrial and academic labs with opportunities to increase purity, yields, and activity.
The 34 µm bead matrix provides high binding capacity (at least 40 mg protein/ml medium), more concentrated samples, higher
efficiency, time savings and increased yield. Furthermore, nickel leakage has been shown to be negligible, which thus helps retain
the activity of the protein and reduce its precipitation. This new medium is also compatible with a wide range of reducing agents,
detergents, denaturants and other additives.
The preparative purification of (His)-tagged recombinant proteins by immobilized metal affinity chromatography (IMAC) is highly
effective. IMAC exploits the ability of the amino acid histidine to bind chelated transition metal ions. His is globally the most used
tag, often found as six histidine residues in series, but it is also present on the surface of many non-modified proteins. Of the metal
ions used in this technique, nickel (Ni
2+
) has been proven to be the most successful. Ni Sepharose High Performance should further
increase the use and reliability of this valuable method of purification and become the first choice medium for the single-step
purification of (His)-tagged recombinant proteins from cellular or cell-free systems.
November saw the opening of the new Large-Scale
Applications Lab at our facility in Piscataway, USA. It
augments the existing applications lab but can accommodate
larger pieces of equipment. Based on a biopharmaceutical
process development lab concept, the new 2 300 sq. ft. lab is
equipped with everything necessary for running all of our
bioprocess technologies, including the our membrane
systems. While not currently in the plans, it would even be
possible to run the OligoProcessTM System. This facility will
complement the new BioProcessTM laboratory currently under
construction in Uppsala, Sweden. A new water system sized
for running filtration systems has been installed with a
5 000L/day capability. For handling large-scale columns two
separate, two-ton hoists (one stationary and one traveling)
enable column assembly and cleaning operations.
The laboratory will be used primarily for proof-of-principle
studies. In the few short weeks after the opening, the lab has
seen customer visits for an FAT (Factory Acceptance Test),
column packing demonstrations and problem-solving.
The new lab was officially opened by Peter Ehrenheim,
president of Protein Separations, at a ribbon-cutting
ceremony and reception for site employees and contractors.
Ni Sepharose High Performance is
available in 25 ml and 100 ml lab
packs, as well as prepacked
HisTrapTM HP 1 ml and 5 ml
columns. HisTrap HP columns can
be used with a syringe, a laboratory
pump or chromatography systems
such as ÄKTATMdesign.
