STREAMLINE Direct column ­ trouble-free
flow from entry to exit
This completely new range of columns simplifies the
distribution and passage of processing liquids, at the same
time as it minimizes the risk of clogging.
Liquid enters the column via a screen-free distribution system
comprising an oscillating arm mechanism. Eliminating the
bottom screen avoids clogging and reduces the risk of
trapping air bubbles. Sample application is also much easier.
At the other end of the column, liquid can exit through a by-
pass rather than the top screen, again eliminating clogging
worries.
Liquid handling as a whole becomes far simpler, the effect of
which will be seen in increased yield, greater reliability and
better economy.
The columns are also easier to clean and sanitize. Microbial
challenge studies have shown that the column design is
effectively sanitized with sodium hydroxide ­ see Application
Note "Sanitizing STREAMLINE Direct 95 column".
New STREAMLINE Direct columns are also longer. We have
extended the maximum sedimented bed height from 30 to
50 cm so you can choose a higher flow velocity with the
same binding capacity. Furthermore, the increased density of
STREAMLINE Direct CST I lets you double the fluid
velocity yet still keep the same bed expansion. This speeds up
capture and reduces the time proteases have to degrade your
target protein. Note that all standard STREAMLINE
adsorbents, including some from our Custom Designed
Media (CDM) service, run in the new column design.
Liquid exits via the net by-pass tube ­ simple, effective and trouble-free.
Simple sample application, especially for liquids with high cell densities
and viscosities.
Simpler purification ­ all in one direction
The whole EBA capture operation now flows in the same
direction ­ in at the bottom of STREAMLINE Direct and out
at the top ­ and steps 2-6 all run in expanded bed mode (see
below).
New STREAMLINE in action ­ a simpler, more direct start to downstream
processing with smoother liquid handling and every step run with upward
flow.
The overall simplicity of STREAMLINE, combined with the
improvements of the new adsorbent and columns, offers a
better, more economical start to downstream processing.
